After whole-mount nuclear (all cells) and fast-green (FG) staining, an E15.5 embryo was cleared & imaged with light-sheet microscopy by Carine Langrez. Only nuclear staining is shown here and the final 3D image was optimised for visualising uncalcified bones (calcified bones can be stained with FG). It was obtained with an in-house signal processing method —developed by Szabolcs Zakany and similar to ‘High Dynamic Range’— that leverages the 12-bit data produced with light-sheet microscopy.

We introduced in 2023 a simple and robust method of whole-mount staining with the ‘Fast Green’ dye that provides unmatched visualisation of 3D collagen network architecture, especially useful in developmental and cancer biology
